Tablet Compression SOP
This Standard Operating Procedure template provides a structured framework to verify that laboratory personnel consistently follow approved procedures when operating the tablet compression machine. It is designed for use by laboratory supervisors during routine walkthrough inspections and SOP compliance evaluations, ensuring adherence to cleaning protocols, equipment setup requirements, and operational best practices throughout the compression process.

How to Use This Checklist
- **Record the inspection details.** Note when the inspection was conducted, who prepared it, and the location.
- **Confirm the purpose and scope statements.** These frame what the procedure covers so the record is self-explanatory.
- **Work through cleaning and preparation.** Verify the compression machine has been cleaned in accordance with the dedicated cleaning SOP using 70% isopropyl alcohol. Check that dust extractors, piping, racks, door frames, and the return filters of the Air Handling Unit have been thoroughly cleaned and verified. Perform a visual inspection to confirm there is no leakage from the equipment. Confirm Quality Control department approval has been obtained and that the 'CLEANED' status label is affixed to the equipment.
- **Walk through equipment setup.** Confirm the 'CLEANED' status label has been removed from the tablet press before operations begin. Check that a product identification label showing the PRODUCT NAME and BATCH DETAILS is affixed to the tablet press. Verify that the appropriate punch set has been issued in accordance with the separate punch management SOP. Confirm machine parameters have been configured according to the tablet specifications in the Batch Manufacturing Record (BMR).
- **Observe the compression operation.** Confirm that tablets produced from the initial machine rotations are discarded before full production of the first batch begins. Check that tablets generated during each batch are collected, packaged, and labeled with the appropriate batch details. Verify the compression process is stopped when the material in the hopper reaches the minimum operating level.
- **Check post-production handling.** Confirm all outer containers have been cleaned and stored with labels indicating complete batch details. Verify punch sets have been removed and cleaned in accordance with the separate punch cleaning SOP.
- **Close out the record.** Sign the inspection, note the finish time, and record the actual start and end dates of the inspection.

Why it matters
Tablet compression sits at the point where a formulated blend becomes a finished dose, so the state of the machine, the tooling, and the surrounding environment directly affects the product. Verifying that cleaning has been completed and approved before setup begins, and that the correct punch set and parameters are in place, keeps production running on approved procedures rather than assumption. Documenting each check creates a record that the process was carried out as written.
What a good inspection covers
A complete walkthrough follows the compression process from end to end: - **Cleaning and preparation** — cleaning of the compression machine with 70% isopropyl alcohol in line with the dedicated cleaning SOP, cleaning and verification of dust extractors, piping, racks, door frames, and the Air Handling Unit return filters, visual inspection for leakage, and Quality Control approval with the 'CLEANED' status label affixed. - **Equipment setup** — removal of the 'CLEANED' label before starting, a product identification label carrying the PRODUCT NAME and BATCH DETAILS, issue of the appropriate punch set per the punch management SOP, and configuration of machine parameters against the tablet specifications in the Batch Manufacturing Record (BMR). - **Operation** — discarding tablets from the initial machine rotations, collecting, packaging, and labeling tablets from each batch, and stopping compression when material in the hopper reaches the minimum operating level. - **Post-production** — cleaning and labeled storage of outer containers, and removal and cleaning of punch sets per the punch cleaning SOP. - **Record details** — who conducted the inspection, the location, the signature, the finish time, and the actual start and end dates.
How the checklist supports the work
The template breaks the process into discrete checks, each answered **Yes**, **No**, or **N/A**. That structure keeps a supervisor moving through cleaning, setup, operation, and post-production in sequence, and it produces a consistent record whether the inspection is routine or part of a broader SOP compliance evaluation. Because several checks refer to separate procedures — cleaning, punch management, punch cleaning — the checklist also makes clear where those documents take over.
